Genrisk-T project: response of the TPC-1 papillary thyroid carcinoma cell line to a range of low to high doses of irradiation

    Abstract

    Our results indicate that TPC-1 cells do not undergo significant cell death but are more prone to cell cycle arrest in response to external X-irradiation. Only a small percentage of irradiated TPC-1 cells underwent apoptosis or necrosis in response to irradiation after 24 and 48 hours. On the other hand, the same cells showed a great decrease in the S phase of the cell cycle, which translates as a decrease in proliferation. Examination of the morphology of irradiated TPC-1 cells showed signs of apoptosis and cellular damage in some of the cells at 24 and 48 hours. In conclusion, our results indicate that irradiation of a human papillary thyroid carcinoma cell line, TPC-1, causes a decrease in cells in the S phase of the cell cycle through activation of the G1/S checkpoint probably caused by an increase in the translational activity of p53 through p21. Apoptosis and necrosis seem not to play as important a role in these cells' response to X-irradiation.
