Considering the interest presented by these materials, a method of synthesis was developed. This method is based on the reduction of the Mannich base obtained by a reaction between uracil, labelled formaldehyde and morpholine . The reduction results in the formation of thymine methyl-14Cwith good yield. Thymidine methyl- 14C is further prepared by means of an enzymatic reaction starting from active thymine. Purification of the products is performed by cristaIIisation for thymine and by paper chromatography with two different eluent systems for thymidine . RadiochemicaI purity of both components is determined by radiochromatography whilst control of Chemical purity and dosimetry are achieved by U.V spectrophotometry .
